Current methods of tissue clearing involve the use of complex organic solvents and many time-consuming steps. These procedures cause a loss of endogenous proteins in the tissue, rendering samples incompatible with further tissue processing or other experimental procedures.
This technology describes a water-based approach for tissue clearing, which renders tissue transparent in only three days and with few steps. The use of organic solvents is minimized, and environmentally-hazardous chemicals are replaced by safer alternatives. Furthermore, this method is compatible with an array of labeling techniques, maintains the integrity of the tissue, and is reversible. As such, this technology may benefit high-resolution 2D or 3D imaging of tissue in a wide range of biomedical fields.
This technology has been validated on whole adult mouse bodies and brains, embryonic spinal cord, and retina.
